Selofen is a medical drug that has a hypnotic and calming effect.

In the structures of neurons of the central nervous system there is a benzodiazepine GABA receptor. In turn, on the alpha substance of the receptor there is omega-1, a subtype of the GABA receptor, which actively reacts with the active substance of Seloven - Zalepton. The hypnotic, calming, anticonvulsant, stress-relieving and anxiety-relieving effect occurs due to changes in the parameters of the membrane chlorine channel.
Pharmacokinetics
Almost complete (at least 71%) absorption of Zalepton in the gastrointestinal tract is noted. After a maximum of one and a half hours, its concentration in the blood plasma will be 37.1 ng / ml. The active substance, seeping into the intercellular space, quickly appears in the tissues of the body, but its concentration will be lower than in the blood serum.
Bioavailability, after biotransformation of Selofen in the liver, will be approximately 30%. Then, pharmacologically inactive metabolites, having undergone transformation to glucuronides, are excreted in the urine.
The half-life of the drug occurs in a fairly short period of time, approximately one hour. This time can be increased to two hours if the patient eats high-fat foods. Also, fatty foods will reduce the concentration of the drug in the blood plasma by half.
Dosing and administration
Selofen can be used in therapy only for adults.
The maximum daily dose should not exceed 10 mg. The second dose of the drug should not be used on the same night as the first.
The duration of treatment is no more than two weeks.
There are some features of taking the drug:
- Patients with mild to moderate liver failure: the daily dose should be a maximum of 5 mg. The drug should not be used at all if this dose does not provide the desired effect. Selofen is contraindicated in patients with severe liver damage.
- Patients with mild or moderate renal impairment do not require dosage adjustment.
- Patients with chronic respiratory failure - the recommended daily dose is 5 mg.
- Elderly patients: the maximum daily dose, due to the high sensitivity of this population category to drugs with a hypnotic effect, should be 5 mg. If the effect is weak, Selofen should not be used.
- Children: The use of Selofen is contraindicated in this group of patients due to insufficient clinical studies.
You should not combine the use of the drug with food intake, because this will significantly delay the absorption of the drug.
Drinking alcohol with Selofen is strictly prohibited.
For maximum effectiveness, the medicine should be taken immediately before bedtime and no earlier than four hours before waking up.
Use Celofen during pregnancy
There is no data on the use of the drug during the gestation period. Therefore, if you are planning or have become pregnant, you should immediately stop treatment. Newborns whose mothers used Selofen in the last weeks of the gestation period or during labor may have low blood pressure or hypothermia. They may also experience mild respiratory depression or develop a "withdrawal syndrome."
Contraindications
Selofen should not be used in therapy:
- if the patient is under eighteen years of age;
- during pregnancy and breastfeeding
- if there is severe liver or respiratory failure;
- if the diagnosis is muscle weakness;
- if the patient's medical history includes a history of sleep apnea;
- in case of individual intolerance to the components of the drug.
Side effects Celofen
When taking the medicine, possible side effects of the body:
- Systemic: malaise, increased sensitivity to light;
- CNS: myalgia, nightmares, anxiety, irritability, psychosis, amnesia, incoherent speech, numbness of the extremities, disorientation.
- Gastrointestinal tract: nausea
- Urogenital system: menstrual irregularities that manifest as pain.
- Immune system: anaphylactic or pseudoanaphylactic reaction
Selofen may cause physical or psychological dependence.
If you stop taking the medication immediately after the symptoms of insomnia have been reduced, sleep problems and withdrawal symptoms may reoccur.
Overdose
If the dose of the drug is exceeded slightly, the patient will feel drowsy and disoriented in space, and lethargy is also possible.
In case of severe overdose, respiratory depression, decreased muscle tone, blood pressure, sometimes coma, and very rarely death may occur.
Life-threatening conditions may occur in patients who consume alcoholic beverages during therapeutic treatment.
Therapeutic treatment is aimed at supporting the body and reducing the symptoms of overdose. You can induce vomiting or wash out the stomach, and also administer a sorbent if the patient is conscious.
The drug Flumazenil shows high efficiency of antidote therapy in animal studies. However, there is no data on the possibility of its use by humans.

Interactions with other drugs
Selofen interacts with some medications:
- Cimetidine is a drug that is a strong inhibitor of liver enzymes. When interacting with the active substance of Selofen, it significantly (up to 85%) increases the concentration of the latter in the blood serum. That is why you should not combine these drugs.
- Erythromcin is a potent selective inhibitor of CYP3A4. It slightly increases the plasma concentration of zaleplon (up to 34%) when interacting, so it does not require dose adjustment. The patient may only feel an increase in the sedative effect of Selofen.
- Rifampicin is a strong liver enzyme inducer. When interacting, it significantly (up to four times) reduces the concentration of Selofen. Also, when prescribing such drugs as Carbamazepine, Phenobarbital, Rifampicin together with Selofen, significantly reduces the effectiveness of the latter.
- Digokin and Warfarin. Selofen can be combined with these drugs with a narrow therapeutic effect.
- Ibuprofen. No interactions.
- Venlafaxine. In a prolonged-action dosage form, it interacts with Selofen. But it does not cause any memory impairment or reduction in psychotropic reactions.
- Opioid analgesics. Increases physical dependence and also enhances the euphoric effect.
- Alcoholic beverages. Interacts and enhances the sedative effect.
- Antihistamines. When taken simultaneously, the sedative effect increases.
Shelf life
The drug can be stored for no more than four years.
