Pharmacodynamics

The active active substance of this drug is a synthetic analogue of the hormone of the adrenal cortex, a non-halogenated methylprednisolone steroid, aceponate.

Methylprednisolone aceponate has lipophilic molecules and penetrates through the lipid membrane of skin cells. When interacting with the protein receptors of the cytoplasm by hydrolysis, 6α-methylprednisolone-17-propionate forms, which enters the cell nucleus and acts as a stimulant for the synthesis of intracellular polypeptides, which in turn inhibit the release of arachidonic acid necessary for the formation of prostaglandins and leukotrienes, inflammatory mediators .

In addition, there is a temporary decrease in the activity of tissue macrophages, fibroblasts, keratinocytes and lymphocytes, which leads to blockade of the release of interleukins - specific mediators of the immune system.

Thus, Advantan removes local inflammation and irritation in the form of itchy rashes, redness and caused by increased proliferation of lichenification cells (thickening of the epidermis).

Pharmacokinetics

After application to the skin in places of inflammation and rashes, Advantan penetrates through the upper layers of the skin, where it is in sufficiently high concentrations for 24 hours. The active substance of the drug manifests its activity only in the foci of inflammation, no more than 2.5% of 6α-methylprednisolone-17-propionate enters the systemic bloodstream. Its inactivation occurs by binding to glucuronic acid in the blood plasma.

Advantan does not accumulate in the body and after 30-32 hours it is completely excreted in the urine.
