Pharmacodynamics

Anastrozole is a non-steroid-type selective element that slows the activity of aromatase. During postmenopause, the formation of estradiol occurs mainly with the participation of estrone produced by peripheral tissues, by transformation from androstenediol. The enzyme aromatase is involved in this process.

Decreased estradiol results in the development of medicinal effects in women with breast cancer. During the postmenopause, the anastrozole component in a dose of 1 g per day leads to a decrease in the values of estradiol by 80%.

The drug does not possess an androgenic, progestagenic, and estrogenic effect and does not affect the release of aldosterone with cortisol in therapeutic portions.

Pharmacokinetics

With the use of drugs inside, it is quickly absorbed inside the gastrointestinal tract. Peak values within the blood plasma are noted after 2 hours (when taken on an empty stomach). Food slightly reduces the absorption rate, without affecting its degree. The synthesis of a substance with a protein in the plasma is 40%. There is no information on the cumulation of the drug.

Exchange processes of LS proceed with the participation of N-dealkylation, glucuronization and hydroxylation. The main product of disintegration is triazole, which does not lead to a slowing down of aromatase activity.

Anastrozole together with its metabolic products is excreted mostly in urine (less than 10% of them have unchanged form), during 72 hours after single use. Half-life is about 40-50 hours.

Dosing and administration

Texol is prescribed for adult women - for oral use, 1 mg once a day.
