Indications Ganaton

The drug Ganaton is prescribed for the activation of gastrointestinal motility in gastroesophageal reflux disease (GERD), chronic gastritis, spasms of the stomach and intestines. Can be used for dyspeptic symptoms (not associated with stomach ulcers and duodenal ulcers); gastralgia, nausea, vomiting, heartburn, flatulence, a decrease or loss of appetite.

Pharmacodynamics

The active substance of the drug Ganaton is the prokinet of tothopride hydrochloride, a blocker (antagonist) of the dopamine receptor (D2) of the gastrointestinal tract. Affecting the D2-receptors, the drug inhibits the function of the catalyst for the hydrolysis of acetylcholine - the enzyme acetylcholinesterase.

This increases the intracellular level of acetylcholine in the nerve receptors of the gastrointestinal tract and provides energy to the cells of the smooth muscles of the digestive organs (due to the unimpeded transformation of ATP into cAMP). As a result, the tone of the muscle fibers of the gastrointestinal tract increases, and the motor system of the digestive tract becomes more active.

Pharmacokinetics

Getting in the gastrointestinal tract, up to 90% of itpotrid hydrochloride is absorbed through the walls of the intestine into the bloodstream; the highest level of the drug in the blood plasma is reached in 40-45 minutes after its administration.

Splinter Ganaton liver enzymes, cumulative effect is absent. Elimination of metabolites occurs with urine with a half-life of about 6 hours.

Dosing and administration

Ganaton tablets are taken orally (whole, one hour before meals) - one tablet (50 mg) three times a day. The maximum permissible daily dose is 150 mg. The standard course of treatment is 14-21 days.

Use Ganaton during pregnancy

Since the safety of the drug Ganaton for pregnant women has not been studied, it is not recommended to use it during pregnancy.

Contraindications

Among the contra-indications of Ganaton are: increased sensitivity to itopride hydrochloride, gastric bleeding, intestinal stenosis, lactation in women, age of 16 years.

Side effects Ganaton

The use of the drug Ganaton can cause such side effects as skin allergic reactions; diarrhea, constipation, abdominal pain, increased dryness in the mouth; headache, sleep disturbances; galactorrhea or gynecomastia (due to an increase in the hormone levels of prolactin in the blood); retention of urination (with a history of hypertrophy of the premenstrual gland).

Overdose

Interactions with other drugs

Ganaton should not be taken concomitantly with anticholinergics, as this reduces its therapeutic effect.

Simultaneous use of this drug and drugs for the treatment of gastric ulcer is allowed.
