The drug Galidor (synonyms - Benzyclan, Benzyclane fumarate, Benzyclamide, Angiocycline) refers to the pharmacological group of myotropic antispasmodics, which, due to a decrease in the muscle tone of the tissues of internal organs and blood vessels, relieve spasms.

Indications of the galidor

The drug Halidor is used in the complex treatment of blood vessel diseases associated with their narrowing (stenosis) or complete blockage (occlusion). These include obliterating atherosclerosis, obliterating endarteritis (thromboangiitis), peripheral forms of nonspecific aorto-arteritis. Galidor is also used in diseases of the vessels of the eyes (obstruction of the central artery of the retina, diabetic angiopathy, etc.)

The drug is prescribed for pathologies of cerebral vessels (acute and chronic forms of cerebral ischemia), with spasms of peripheral vessels characteristic of acrocinosis, idiopathic Reynaud's syndrome and fever.

Indications for the use of Galidor are diseases of the gastrointestinal tract and urinary system, which are accompanied by spasms of internal organs: gastroenteritis, peptic ulcer and duodenal ulcer, cholecystitis, cholelithiasis, colitis, spastic dissenosis of the large intestine, frequent painful urge to defecate ( tenesmus), flatulence after operations, spasms and tenesmus of the bladder.

Pharmacodynamics

The therapeutic effect of the preparation Halidor is based on the biochemical action of its active substance benzyclane (N, N-Dimethyl-3 - [[1- (phenylmethyl) cycloheptyl] oxy] -1-propanamine), which is capable of changing the ion balance in the membranes of smooth muscle cells.

As a result, activation of the cell enzyme ATP-pyrophosphate lyase with the simultaneous blockade of membrane proteins - adenosine receptors - is triggered. The result of all biochemical reactions is the reduction of cell contractions of the tissue of the visceral musculature (the muscles of the internal organs). Thanks to this there is a relaxation of muscles, a decrease in the tone of blood vessels and the removal of spasm.

Pharmacokinetics

Getting into the gastrointestinal cycle (when taking the drug inside), the active substance of the drug Galidor is rapidly absorbed, and its maximum concentration in blood plasma is reached on average after three hours. About 40% of the drug binds to plasma proteins, up to 30% for etritocytes, 10% for platelets.

After passing through the liver, the bioavailability of Halidor (after oral administration) is a maximum of 35%.

Benzyclone is metabolized in the liver; the final decomposition products are excreted in the urine (the half-life is between 6 and 10 hours). The speed of cleansing plasma from the drug substance (total clearance) is 40 liters per hour, the renal clearance is less than 1 liter per hour.

Dosing and administration

Halidor in the form of tablets in the pathology of the vessels is taken one tablet (100 mg) three times a day. The duration of treatment on average is two months, during which the doctor can reduce the dosage to one tablet twice a day.

To remove persistent spasms of internal organs, Halidor is prescribed 1-2 tablets (100-200 mg) - once a day; for maintenance therapy - on a tablet three times a day. The maximum daily dose of this drug is 400 mg, the duration of the course of therapy is no more than 1-2 months.

The method of application of Galidor in the form of 2.5% solution: intramuscularly (if necessary, intravenously - slowly) 1-2 ml per injection. In diseases of blood vessels, the solution can be infused (dropwise) into the vein (4 ml of the drug are diluted with 100-200 ml of isotonic NaCl solution).

When spasms of internal organs the drug is injected into a vein (with isotonic solution) or deep into the muscle. The standard course of treatment lasts no more than three weeks.

Use of the galidor during pregnancy

The use of Halidor during pregnancy is contraindicated. If Galidor is prescribed, lactating women should stop breastfeeding while taking the drug.

Contraindications

Among the contraindications to the use of the drug Galidor are: severe forms of respiratory, renal, hepatic and decompensated heart failure, myocardial infarction, ventricular tachycardia, epilepsy, hemorrhagic stroke, craniocerebral trauma (suffered during the last year), age to 18 years.

Side effects of the galidor

Overdose

Overdose of Galidor is expressed in the increase in heart rate, blood pressure decrease, spontaneous urination, drowsiness or anxiety; there may be a collapse, convulsive seizures. With such symptoms, a gastric lavage should be done.

Interactions with other drugs

Reception Galidor with drugs that reduce the content of potassium (diuretic, laxatives), enhances their effect. Admission with cardiac glycosides (digitalis preparations) can cause cardiac arrhythmias. The simultaneous use of Galidor with drugs for the treatment of cardiac arrhythmia or arterial hypertension (non-selective and selective beta-blockers) can cause tachycardia.

The interaction of Galidor with sedative (soothing) drugs and means for anesthesia is expressed in the intensification of their action on the central nervous system. And the use of Galidor and the intake of aspirin (acetylsalicylic acid) can disrupt aggregation (gluing) of platelets and lead to bleeding.
