Pharmacodynamics

Tocopherol is a fat-soluble vitamin that has a pronounced radioprotective and at the same time antioxidant effect, participates in the processes of heme and protein biosynthesis, cell proliferation and other important processes of cellular metabolism. The component improves oxygen saturation of tissues, has an angioprotective effect, affecting vascular permeability and tone by stimulating the processes of formation of new capillaries.

The immunomodulatory activity of tocopherol develops with stimulation of humoral and T-cell immunity.

This vitamin is essential for healthy reproductive function: conception, fetal development, and the formation and functioning of the reproductive system.

Tocopherol deficiency causes myocardial and skeletal muscle dystrophy, as well as hypotension, reduces the strength of capillaries and increases their fragility, and also leads to degeneration of photoreceptors, which causes visual impairment. Along with this, there is a weakening of reproductive activity in men, and in women - a disorder of the menstrual cycle, as well as an increased tendency to miscarriages.

A deficiency of tocopherol may result in hemolytic jaundice in newborns, as well as steatorrhea or malabsorption syndrome.

Pharmacokinetics

After intestinal absorption, the main part of the drug penetrates into the blood with lymph and undergoes rapid distribution within the body tissues, during which mainly accumulation occurs within the muscles, liver and fatty tissues. Maximum drug levels are noted within the pituitary gland with the myocardium, adrenal glands and sex glands.

The major part of Enata 400 is excreted in urine, and the remainder in bile.

Overdose

Administration of large doses of tocopherol (0.4-0.8 g per day over a long period of time) may cause diarrhea, visual disturbances, headaches, abdominal cramps or nausea, as well as dizziness, extreme weakness or fatigue. Increased CPK values, serum triglycerides and cholesterol levels may be observed, as well as increased androgen and estrogen values in the urine.

Extremely large doses (over 0.8 g, over a long period of time) may cause bleeding in people with vitamin K deficiency. It may also disrupt the metabolism of thyroid hormones and increase the risk of thromboembolism with thrombophlebitis in people with severe sensitivity.

To eliminate the disorder, tocopherol should be excreted from the body and then symptomatic measures should be taken.

Interactions with other drugs

Tocopherol is prohibited to be used in combination with silver or iron substances, as well as drugs with an alkaline environment (trisamine and sodium bicarbonate) and indirect anticoagulants (neodicoumarin with dicoumarin).

The drug potentiates the activity of SSAIDs and NSAIDs (ibuprofen with sodium diclofenac and prednisolone), and also reduces the toxicity of SG agents (digoxin with digitoxin), calciferol and retinol.

Tocopherol and its metabolic products have an antagonistic effect relative to vitamin K.

The drug increases the therapeutic effectiveness of anticonvulsants in people suffering from epilepsy.

Colestipol with cholestyramine and mineral oils reduce the absorption of tocopherol.

Using large amounts of tocopherol can lead to a decrease in the body's retinol stores.
